Functional Genomics Facility (FGF)

The Functional Genomics Facility (FGF) aims to assist researchers in a variety of projects. This recently established, fully equipped facility has full-time staff and can undertake the following:

DNA Sequence Analysis:

The facility is equipped with an ABI 3730 capillary based sequencer. The facility provides:

  • Standard sequencing from a variety of templates (plasmids, PCR products, etc.)
  • Hairpin sequencing (shRNA and miRNA encoding DNAs)
  • Bisulfite sequencing (methylation analysis)

Quantitative PCR

SYBR green and Taqman quantitative PCR analyses are performed with ABI 7900HT real-time PCR machine (96 and 384 well plate) and Stratagene MX3000 QPCR machine (96 well plate). Services include:

  • Preset plate run (96 or 384 well plates)
  • Sample preparation and analysis
  • Assay design and optimization

Construction of Retroviral and Lentiviral Vectors

  • FGF will generate customized lentiviral and retroviral expression vectors, including:
  • Vectors expressing protein of interest under control of various promoters.
  • shRNA expressing vectors, allowing for drug- and fluorescent protein-based selection of transduced cells
  • Reporter vectors, allowing to analyze activity of the promoters to monitor a variety of luminescent (luciferases) and fluorescent reporter proteins

Custom Production of Recombinant Lentiviruses and Retroviruses

The Facility is providing custom production of recombinant lentiviruses and retroviruses. The services include:

  • Lentiviral vector production on a small (20 ml), standard (200 ml) and large (up to 1 L) scale
  • Retroviral vector production on a small (20 ml), standard (200 ml) and large (up to 1 L) scale
  • Virus concentration (preparation of concentrated virus stocks, with titers up to 1010 MOI)
  • Viral vector titering (estimation of real titers of retroviruses and lentiviruses based on QPCR with virus specific primers)

Chemical Library Screening

The facility provides access to ChemBridge and MicroSource small molecules libraries. The facility designs and performs a variety of cell-based assays, using full libraries or custom subsets. Assays can be fully automated, which allows for high-throughput screening of our diversified libraries. The Sciclone liquid handling system is linked to an incubator, plate washer and Synergy HT plate reader through a Twister II robot with multi-plate stacker. Two additional Rapid Plate liquid handling systems are also available and can be integrated or used as a
stand-alone.

siRNA GENE SILENCING

The FGF provides access to Qiagen Human Whole Genome siRNA set. The facility performs high-throughput screenings with the entire collection or custom-specified siRNA subsets. The services include:

  • Optimization of siRNA transfections
  • Preparation of custom siRNA subsets
  • High-throughput screening with the entire siRNA library or its subsets

Full Length Human cDNA Clones

The facility will provide cDNA clones from MGC and ORF 1 and 2 collections. (Open Biosystems)
